The 1999–2000 Penn State Lady Lions basketball team represented Pennsylvania State University during the 1999–2000 NCAA Division I women's basketball season.

The Lady Lions, led by 20th year head coach Rene Portland, played their home games at the Bryce Jordan Center and were members of the Big Ten Conference.

3 seed Purdue in the 2000 Big Ten Conference women's basketball tournament.

They were invited to the 2000 NCAA Division I women's basketball tournament where they reached the first Final Four in program history before losing in the National semifinals to eventual champion UConn.
