Bob and Mike Bryan defeated Rohan Bopanna and Aisam-ul-Haq Qureshi in the final, 7–6(7–5), 7–6(7–4) to win the men's doubles tennis title at the 2010 US Open.

The pair did not lose a set during the tournament.

Lukáš Dlouhý and Leander Paes were the defending champions, but lost in the first round to Martin Damm and Filip Polášek.
